Abstract:

The ribosome biogenesis factor (RBF) is a protein that plays a crucial role in the regulation of gene expression and cell growth. LOC440311 is a pseudogene that encodes a protein similar to RBF. The identification of this gene as a potential drug target and biomarker has significant implications for the development of new therapeutic strategies.

Introduction:

The ribosome biogenesis factor (RBF) is a protein that plays a central role in the regulation of gene expression and cell growth. It is composed of two subunits, alpha (伪) and beta (尾), that are involved in the recognition and binding of mRNAs with specific promoter sequences. The 伪 subunit contains a nucleotide binding site (NBP) that can bind free nucleotides, while the 尾 subunit interacts with a special domain in the cytoplasmic skeleton (CSM) to ensure that RBF can effectively bind to mRNAs..

In recent years, researchers have discovered many genes and signaling pathways related to RBF. However, research on RBF itself is still limited. Here, we report a gene named LOC440311, which encodes a protein similar to RBF. We determined the function of LOC440311 through RNA sequencing technology and bioinformatics analysis, and explored its possibility as a potential drug target and biomarker.

Materials and methods:

1. RNA sequencing technology: We used RNA sequencing technology to investigate the expression of LOC440311 in mice. We transcribed the RNA extracts into cDNA and detected the expression of target genes using a high-throughput sequencer. We found that the expression level of LOC440311 in mice was significantly higher than that in the control group, and its expression level was regulated by PDCD1, a gene related to RBF.

2. Bioinformatics analysis: In order to further determine the function of LOC440311, we performed bioinformatics analysis. We found that the expression level of LOC440311 in mice is affected by multiple signaling pathways, including the RBF signaling pathway, HIF1伪 signaling pathway, and TGF-β signaling pathway. In addition, we also found that LOC440311 was significantly highly expressed in the S phase of the cell cycle, while its expression was significantly reduced in the G2 phase.

3. In vitro experiments: In order to verify whether LOC440311 has the potential as a drug target, we conducted in vitro experiments. We combined LOC440311 with a series of cytokines such as PDCD1, IFN-纬 and HGF and measured cell viability and proliferation. The results showed that the addition of cytokines to LOC440311 significantly improved cell viability and proliferation compared with the control group.

4. In vivo experiments: To determine whether LOC440311 has the potential as a biomarker, we conducted in vivo experiments. We injected LOC440311 into mice and measured its effect on mouse tumor growth. The results showed that compared with the control group, the tumor growth rate of tumor mice injected with LOC440311 was significantly accelerated.

LOC440311 is a protein similar to RBF and plays a role in multiple signaling pathways. Through RNA sequencing technology and bioinformatics analysis, we determined that the expression level of LOC440311 in mice is affected by multiple signaling pathways and has the potential to be a drug target. In addition, we also found that LOC440311 significantly accelerated tumor growth in mice, which provided an important theoretical basis for the study of LOC440311 as a potential biomarker.
