TRAPPC1: A Protein Targeted for Drug Development and Tumor Biology

Trafficking Protein Particle Complex Subunit 1 (TRAPPC1) is a protein that plays a crucial role in the transport of proteins from the endoplasmic reticulum (ER) to various extracellular membrane spaces (extramembrane space). biological function, and is a popular target in drug development.

TRAPPC1 is a ~200kDa protein that belongs to the glycoprotein family. It is composed of two domains: N-terminal 伪-helix and C-terminal 尾-coil. The N-terminal 伪-helix of TRAPPC1 consists of 150 amino acids and is a key region for its function in membrane transport.

The function of TRAPPC1 is mainly reflected in its transport mechanism. In cells, TRAPPC1 is transported to the Golgi apparatus (Gastrointestinal mucosa, GIM) by forming a ~200 nm particle, and then binds to transfer RNA (tRNA) through GIM, and transports tRNA to the target cell membrane in the cytoplasm. In GIM, the N-terminal 伪-helix of TRAPPC1 pairs with the 伪-helix in GIM and interacts with the 尾-coil in GIM through non-covalent interactions. Subsequently, TRAPPC1 moves to the cell membrane along the ribosomes in the cytoplasm by binding to the transfer RNA. On the cell membrane, TRAPPC1 pairs with proteins on the cell membrane through its C-terminal 尾 coil and transports them from the cell membrane to the outside of the cell.

TRAPPC1 is widely used in drug development. Since TRAPPC1 plays an important role in drug transport, it is used as a target for drugs. Some studies have shown that inhibitors of TRAPPC1 can inhibit tumor growth and metastasis and improve patients' quality of life. Currently, there are some TRAPPC1 inhibitors on the market used in clinical trials, such as ALX148, ALX214, and ALX278.

TRAPPC1 is also a potential biomarker. Since TRAPPC1 plays an important biological function in cell membrane transport, its expression level can reflect the activity of cell membrane transport. Some studies have shown that the expression level of TRAPPC1 is positively correlated with tumor invasion and metastasis. Therefore, TRAPPC1 can serve as a potential tumor biomarker for predicting tumor growth and metastasis.

TRAPPC1 has important application value in drug development and tumor biology. With the continuous advancement of technology, TRAPPC1 inhibitors will bring more hope to clinical treatment.
